This work assesses the antibacterial activity of plumbagin (5-hydroxy-2-methylnaphthalene-1,4-dione) and of methanol, chloroform and aqueous extracts of Plumbago zeylanica L. root against various pathogenic bacteria, and the minimum inhibitory concentrations (MICs). Plumbagin and chloroform extracts of Plumbago zeylanica L. root showed antibacterial activity against Escherichia coli, Salmonella typhi and Staphylococcus aureus. Inhibition against Klebsiella pneumoniae, Serratia marcescens and Bacillus subtilis was moderate, and lower against Proteus vulgaris and Pseudomonas aeruginosa. The methanolic extract exhibited moderate activity and the aqueous extract weak activity against the bacterial strains as assessed by disc diffusion assays. The bioactive compound plumbagin and extract of Plumbago zeylanica root show a wide spectrum of antibacterial activity. The compound shows promise as a new drug for various bacterial infectious diseases. |

Plumbago species (Family-Plumbaginaceae) are one of the most important medicinal plants which are used for anti-inflammatory, antimicrobial diseases. Our work involves the study of anti inflammatory and cytotoxic effects of Plumbago zeylanica. The root of P. zeylanica extracted with methanol was used for determining the anti inflammatory effects. The methanolic extracts at 300 and 500 mg/kg produced 31.03 and 60.3% inhibition of acute inflammation, respectively, in Carrageenin induced raw paw oedema confirming that P. zeylanica roots are effective against acute inflammation. For the evaluation of cytotoxicity, the crude dichloromethane extract was subjected to silica gel column chromatography and 120 fractions were collected. Their structures were elucidated with the help of spectroscopic techniques. High performance liquid chromatography (HPLC) was performed to determine the purity of gugultetrol-18-ferrulate in crude extract and the structure of betasitosterol and gugultetrol-18-ferrulate was identified using nuclear magnetic resonance spectroscopy analysis ( 1H and 13C NMR), Infra red and mass spectroscopy. The lethal concentration (LC50) value was observed for crude extract, betasitosterol, gugultetrol-18-ferrulate and it was found to be 90, 75 and 65 ppm, respectively. The use of Plumbago species as an effective anti inflammatory agent and its cytotoxic effects have been ascertained and proved. |
| 3 | Antioxidant properties of Plumbago zeylanica, an Indian medicinal plant and its active ingredient, plumbagin | Plumbago zeylanica (known as 'Chitrak') is a useful Indian medicinal plant. The root of the plant and its constituents are credited with potential therapeutic properties including anti-atherogenic, cardiotonic, hepatoprotective and neuroprotective properties. To examine possible mechanisms of action of P. zeylanica (Chitrak), in relation to its reported beneficial properties, antioxidant effects of the aqueous/alcoholic extracts of root, corresponding to medicinal preparations, and the active ingredient, plumbagin, were studied. Methods used included: ferric reducing/antioxidant power (FRAP), radical scavenging of 1,1-diphenyl-2-picryl hydrazyl (DPPH) and 2,2'-azobis-3-ethylbenzthiazoline-6-sulfonic acid (ABTS), lipid peroxidation in rat liver mitochondria induced by different agents, and estimating phenolic and flavonoid content. In FRAP/DPPH assays, boiled ethanolic extracts were the most effective, while in the ABTS assay boiled aqueous extracts were the most efficient. These extracts also significantly inhibited lipid peroxidation induced by cumene hydroperoxide, ascorbate-Fe2+ and peroxynitrite and contained high amounts of polyphenols and flavonoids. To examine the mechanisms of action in detail, antioxidant and pulse radiolysis studies with plumbagin were conducted. The hydroxyl (•OH), alkyl peroxyl (CCl3OO•), linoleic acid peroxyl (LOO•), and glutathiyl (GS•) radicals generate a phenoxyl radical upon reaction with plumbagin. The bimolecular rate constants were: •OH, 2.03 x 109 dm3mol–1s–1; CCl3OO•, 1.1 x 109 dm3mol–1s–1; LOO•, 6.7 x 107 dm3mol–1s–1; and GS•, 8.8 x 108 dm3mol–1s–1. In conclusion, our studies reveal that extracts of P. zeylanica and its active ingredient plumbagin have significant antioxidant abilities that may possibly explain some of the reported therapeutic effects. |

The present review deals with chemical compounds, medicinal properties, biological activities and pharmacological effects of Plumbago zeylanica. Plumbago zeylanica (Linn.) belongs to family Plumbaginaceae, commonly known as Chitrak is one of the medicinal plants used in the Indian traditional system of medicine. Traditionally P. zeylanica is used as a stimulant digestant, expectorant, laxative and in the treatment of muscular pain and rheumatic diseases. Some parts of P. zeylanica are used in various pharmacological activities. The different parts of P. zeylanica are used for various ethnomedicinal purposes and investigations have been carried out on different chemical compounds such as plumbagin and other compounds of this plant. Its biological activities like antibacterial, antimycotic, antiviral, antiplasmodial, leishmanicidal, trypanocidal and anticarcinogenic have been studied along with pharmacological effects of it. |
| 5 | Heneicosane—A novel microbicidal bioactive alkane identified from Plumbago zeylanica L | Today the quick advancement of multi drug resistance in microorganisms is expanding worldwide and the related medical issues for the treatment of infectious diseases that researchers claim could return to the stage of the pre-antibiotic era. Medicinal plants possess a potential holistic role for the provision of drugs to cure microbial infections. The present work, has aimed to isolate and analyze the new bioactive molecule and to test their antimicrobial activities from the Plumbago zeylanica L leaf extracts. For analyzing the phytochemistry and microbicidal activities of the herb, extracts with various solvents have been used. The various crude leaf extracts with antimicrobial activity was treated against pathogenic microorganisms such as two Gram positive, two Gram negative, one acid fast staining bacteria and one fungus using agar well method. New bioactive molecule was isolated via GC–MS, TLC, HPLC, FTIR and NMR spectrum. Current study had used ethyl acetate leaf extract and exhibited excellent antimicrobial activity compared with other extracts and conventional drugs. The ethyl acetate extract was analyzed by GC–MS which was detected in fifteen compounds, followed by TLC analysis to elucidate the ten compounds with ten fractions. Among the active fractions, a single compound was detected using HPLC analysis. The identified bioactive molecule Heneicosane was used for measuring the antimicrobial activity. Depending on the results obtained, it was verified that heneicosane exhibited excellent antimicrobial activity against Streptococcus pneumoniae 31 ± 0.64 mm and Aspergillus fumigatus 29 ± 0.86 mm respectively at 10 μg/ml concentrations. Hence this study derives its novelty in isolating the bioactive molecule with pronounced microbicidal role from the herbal source mentioned. |
| 6 | Potential anti-inflammatory activity of plumbago zeylanica | Oral administration of dichloromethane extract of Plumbago zeylanica (DMEPZ) shown a significant (p<0.05) dose-dependent protection against carrageenan-induced paw edema. At 1st hr, P. zeylanica shown an inhibition effect of edema in the different doses of 250 mg/kg and 500 mg/kg were found to be 28.57 and 31.79%, respectively. At 3rd hrs, the paw edema inhibition was found to be 30.70 and 40.15%, respectively. Diclofenac (25 mg/kg) had effect of 34.10 and 41.73% (p<0.001) inhibition of paw edema at in 1 and 3 hrs. P. zeylanica 500 mg/kg showed percentage inhibition of wet and dry cotton pellet granuloma in rats 55.84% and 47.92%, respectively. Thus, the present study revealed that the DMEPZ offered significant protection against inflammation. |
